A reagent of  phenol and guanidine-salt inhibits RNase activity, and so the reagent maintains integrity of the RNA during  sample homogenization or lysis. A biological sample is homogenized or lysed in R&A-BLUE™ and the  homogenate is separated  into aqueous and organic phases by chloroform addition and centrifugation. RNA  remains exclusively in the aqueous phase, DNA in the inter phase and proteins in the organic phase. RNA is precipitated from the  aqueous phase by addition of 2-propanol, washed with ethanol and solubilized. R&A-BLUE™ Total RNA Extraction Kit performs well with large or small amount of tissue or cells and many samples can be simultaneously extracted. This is one of the most effective methods for isolating total RNA and  can be completed in only 1 hour starting with fresh tissue or cells. The procedure is very effective for isolating  RNA molecules of all types. The isolated RNA is intact with little or no contaminating genomic DNA. This RNA  can be used for Northern blot, cDNAsynthesis, in vitro translation, cloning and RT-PCR.

Plant accumulates variety of metabolites through photosynthesis and those metabolites would  be an inhibitor which has an effect on extraction of nucleic acid. To remove this inhibitor, high concentrationchaotrophic salt or phenol should be used under lysis step. However, for  rice, barley, wheat and seed containing a lot of starch or bean, green gram, pea and seed containing a lot of protein as well as soil, if alcohol is added for binding, too much precipitation  occurs and also the nucleic acid is precipitated (co-precipitation), that brings the sample including less RNA.
